The most important part is tuning the antenna. A badly tuned antenna can melt output transistors in a CB radio.

This video should help. If you don't have a watt meter it may be best to take it to a shop but it can be done without as shown in these videos hope they help.

I don't have experience with CB gear (only ham), but I would expect that if you buy a new commercially-made radio, it shouldn't require any internal tuning (for modulation, etc.) - all that should be done at the factory...(?) As suggested, the antenna is the thing that requires tuning, since that will vary considerably, depending how and where it's mounted.
First, make sure the antenna is properly grounded (generally via the mount). Second, find somebody with an SWR meter... and tune the antenna in a wide open area, with doors closed. The radio itself should be factory tuned. But even with a well tuned antenna, don't expect much... it's a CB after all.
